| Looking to reclaim his place as King of Hell, Crowley asks Dean for help finding “The First Blade,” the only blade that can kill Abbadon. Knowing he owes Crowley a favor for helping Sam and also wanting Abaddon dead himself, Dean reluctantly agrees and the two set off on a road trip. However, Dean and Crowley are completely unprepared when they find out who has the blade — Cain, the first son. Meanwhile, Sam is back at the bunker with Castiel, who notices there is some leftover grace inside Sam. If they can extract it, they can use the grace to track Gadreel. The only problem is the procedure may do permanent damage to Sam... |

- There is one weapon which can kill a Knight of Hell - the "First Blade", which Cain used to kill Abel.
- Cain killed his brother out of love, not hate, to stop him becoming Lucifer's protégé.
- Cain created the Knights of Hell and he destroyed them, not the archangels.
- The "Mark of Cain" as well as the First Blade is needed to kill Knights and Cain can transfer it to someone "worthy" - Dean!

- The episode's title "First Born" is likely a reference to the fact that Cain and Dean are both first born sons. Cain killed his brother, Abel (presumably to save his soul from damnation), and Dean was forced to lie to his brother to save him
- Jensen did his own stunts in the main fight scene in this episode

- When Crowley initially approaches Dean, he jokingly asks if they have time "for a montage", a movie device that shows brief clips of a character (or characters) against a musical background
- In colloquial language, "Droog" refers to a fellow gang member; Crowley uses the term to describe a demon underling who works for him
- In the bible's Book of Genesis, Cain did kill his brother, thereby committing the world's first murder; but Cain killed Abel out of jealousy, not to save his soul. In the original bible story, the Lord gave Cain a Mark so that other people he encountered wouldn't kill him because of what he'd done
- Crowley says to Dean "Didn't they teach note-taking at Hunters Hogwarts?", referencing Hogwarts School of Magic in J.K. Rowling's "Harry Potter" novels
- Dean berates Crowley, saying "...like your heart grew three sizes". This refers to the fictional character The Grinch from the Dr. Seuss children's book (and later an animated film) How the Grinch Stole Christmas. In the book and film, the Grinch is initially selfish and mean-spirited because his heart was too small, but after he witnesses acts of selflessness and kindness, his heart grown three sizes and he changes his ways
